PS 1145 is a specific IKK inhibitor (IC50 = 88 nM), displaying no inhibitory activity against a panel of other tested (in vitro) kinases (PKA, PKC, CK2, MEK1, p38, Lck, Jnk, ERK2, EGFR, IC50 > 100 �M). Treatment of multiple myeloma cells with PS 1145 results in a complete inhibition of I?B? phosphorylation triggered by TNF?, which in turn prevents DNA binding of NF-?B. Furthermore, PS 1145 induced caspase 3/7-dependent apoptosis and significantly sensitized prostate carcinoma cells to apoptosis induced by tumor necrosis factor alpha. Cells cultured in presence of PS 1145 showed an altered proliferation profile, caused by suppressed expression levels of cyclins (D1 and D2).
