The heatstable protein kinase inhibitor PKI is a key role player for the regulation of PKA activity and for localizing the catalytic subunit of PKAin the cell, also working as a nuclear scavenger. This relatively small protein (77 amino acids) is extremely selective for PKA and binds with very high affinity to the catalytic subunit of PKA (Ki = 0.5nM). It favors the nuclear export of the catalytic subunit thereby blocking the cAMP response element regulated gene expression. The recombinant heat-stable protein kinase inhibitor PKI supplied by Biaffin is a 8 kDa monomeric protein without fusion tag suitable for inhibition studies with PKA catalytic subunit. The amino acid sequence of the rabbit PKI shows 100% identity to the human PKI protein.
