PF 4708671 is a cell-permeable specific inhibitor of ribosomal protein S6 kinase 1 (Ki of 20 nM, IC50 of 160 nM(in vitro)). The compound is highly selective for S6K1 and exhibits no significant inhibition for 90 tested protein and lipid kinases, showing a 400-fold less potency against S6K2. In HEK-293 cells, PF-4708671 inhibits S6K1 activity upon stimulation with IGF1 and PMA. Furthermore, PF 4708671 induces phosphorylation of the T-loop and the hydrophobic motif of S6K1, an effect that is dependent upon mTORC1.
