AZD 8055 is potent ATP-competitive inhibitor of mTOR kinase (Ki = 1.3 nM) displaying an at least 1,000-fold lower potency against closely related class I and class III PI3K and the PIKK family members ATM and DNA-PK. Phosphorylation of Akt on the mTORC2 site was inhibited upon treatment of different cell lines with AZD 8055. Furthermore, the compound inhibited also mTORC1 signaling, as determined by a complete loss of phosphorylation of 4E-BP, which in turn potently inhibits cap-dependent translation. AZD 8055 potently inhibits proliferation and induces autophagy and cell death in cancer cells and is still in clinical phase 1 trials.
