Elk-1 belongs to the ETS family of transcription factors controlling various cellular functions. Their characteristic property is the ability to form a ternary nucleoprotein complex with the serum response factor (SRF) dimer. Elk-1 ist structured by an ETS domain that binds to the the 5-GGAA/T-3 core motif of DNA, an activation domain targeted by kinases, a repressor domain that dampens the activity of the activation and a domain required for ternary complex formation. The transcriptional activity of Elk-1 is stimulated by phosphorylation by MAP kinases (ERK, JNK and p38) and can modulate transcription of genes by interaction with the serum response element (SRE) of the c-fos promoter.
