Pyridoxal kinase is an enzyme involved in vitamin B6 metabolism. Since mammals cannot synthesize pyridoxal-5-phosphate (PLP), a cofactor for enzymes primarily involved in amino acids and sugar metabolism, dietary precursors such as pyridoxal, pyridoxamine and pyridoxine (classified collectively as vitamin B6) are essential. PDXK catalyzes the phosphorylation of vitamin B6 in the presence of ATP and divalent cations (Mg2+, Zn2+). In recent studies it becomes evident that genetic variations of PDXK are associated with Parkinson disease, since PLP is involved in the biosynthesis of dopamine, a neurotransmitter linked to the disease characteristics such as motor and movement disorders.
