Protein kinase C (PKC) is a family of serine-threonine kinases, which are classified into three major groups: classical (alpha, beta and gamma), novel (delta, epsilon, eta, and theta) and atypical (mu, xi and iota). In contrast to classical isoforms PKC zeta exhibits a kinase activity which is independent of calcium and diacylglycerol but not of phosphatidylserine. Furthermore, it is insensitive to typical PKC inhibitors. Unlike the classical PKC isoenzymes, it has only a single zinc finger module. The ceramide binding domain (C20zeta) in the carboxyl terminus of PKC zeta has emerged as important binding partner for ceramide affecting the formation of junctional complexes in epithelial cells. Furthermore, pre-activation of PKC xi by ceramide or ceramide analogs sensitizes the enzyme to inhibition by PAR-4, resulting in a down-regulation of NF-kappaB-dependent cell survival.
