The Basic Cytotoxicity Test kit contains two fluorescent reagents. The first, 5-(and-6)carboxyfluorescein diacetate, succinimidyl ester (CFSE), is an amine-reactive, green fluorescing cell protein/membrane stain used to label/identify target cells in your cytotoxicity assay. The second reagent is 7-aminoactinomycin D (7-AAD), a red live/dead viability stain, used to identify the killed/dead target cells present in the cytotoxicity assay samples. Using this BCT kit, all of the target cells are initially labeled with CFSE. Then unstained immune effector cells, cytotoxic drug compounds, or antibodies + complement can be added and incubated for the desired length of time. Following incubation, the 7-AAD is added to stain all of the dead cells red. The cells are now ready for analysis, typically by the use of flow cytometry. The percentage of cell cytotoxicity is calculated by counting the number of dead target cells (that are stained both green and red) and dividing by the total number of target cells in the sample (all green cells).
