A novel tGFP-Glucocorticoid Receptor cell line has been developed through stable transfection to observe the dynamics of the hormone/receptor complex in living cells and to assay for compounds inducing tGFP-hGR nuclear translocation and formation of nuclear foci. HEK293/tGFP-hGR cell line was obtained by transfection of an expression vector for a fusion protein of turboGFP and human GR, as well as G418 into HEK cells with Evrogen TurboGFP. Single cells with strong green fluorescence were selected by flow cytometry, and allowed to expand. These cells constitutively express the tGFP-hGR fusion protein.

Innoprot GR nuclear translocation Assay has been designed to assay compounds or analyze stimuli for their ability to modulate glucocorticoid receptor nuclear translocation process and quantifying the fluorescence distribution inside the cells.

This highly reproducible assay allows monitoring glucocorticoid receptor nuclear translocation process in High Content Analysis and fluorescence microscope applications.

Materials Provided
