The PTEN gene (aka MMAC1 and TEP1) is a candidate tumor suppressor gene. Mutations of PTEN are implicated in sporadic brain cancer, breast, kidney and 60-80% of prostate cancers. PTEN functions primarily as a lipid phosphatase to regulate signal transduction pathways; a key target is phosphatidylinositol 3,4,5 trisphosphate, a signalling inositol phospholipid generated by PI3K activity.
